Abstract

Molecular markers are important tools in studying genetic traits and can help identify the relationship between genes and key economic traits, such as sweetness in pineapples. Varieties like MD2 and Tradt Si Thong are popular for fresh consumption due to their sweetness and good taste. This study aimed to validate whether molecular markers are related to sweetness in fresh pineapples from a population derived from a cross between MD2 and Tradt Si Thong, using markers umc1594 and umc2144. The experimental process began with the collection of 100 young pineapple leaf samples for DNA extraction using the CTAB method. The quality and quantity of the extracted DNA were then assessed using a Nanodrop machine. DNA was subsequently amplified by PCR, and the suitability of the molecular markers was checked using gel electrophoresis. The results showed that the molecular markers umc1594 and umc2144 did not amplify DNA by PCR, as no DNA bands were observed on the agarose gel. Therefore, these markers could not be used to identify the sweetness trait. However, the same technique could be applied in combination with other molecular markers that are linked to sweetness, increasing the likelihood of identifying suitable markers.
